Beras hitam (Oryza sativa L.indica) merupakan salah satu beras berpigmen yang kaya akan senyawa fenolik dan berpotensi sebagai antioksidan alami. Penelitian ini bertujuan menentukan kadar total fenolik pada fraksi air hidrolisis HCl dari ekstrak etanol beras hitam. Penelitian ini dilakukan pada bulan meret-juli 2025 dilaboratorium Kimia Farmasi. sampel berasal dari Kabupaten Slamet Jawa Tengan Menggunakan. litian ini mengunakan metode Eksperimental dengan analisis kuantitatif dan kualitatif. Hasil ekstraksi menunjukkan rendemen 10,15%. skrining fitokimia mengidentifikasi alkaloid, flavomoit, tanin, fenolik, saponin, steroid & terpenoid. Fraksinasi menghasilkan fraksi air yang kemudian dihidrolisis dengan HCl untuk membebaskan glikon. Penetapan kadar total fenolik menggunakan metode Folin–Ciocalteu dengan spektrofotometri UV-Vis pada panjang gelombang 730 nm, menggunakan asam galat sebagai standar. Hasil penelitian menunjukkan kadar fenolik total fraksi air sebesar 73,402 ± 0,173 mg GAE/g %, sedangkan fraksi etil asetat setelah hidrolisis meningkat menjadi 91,72 ± 0,420 mg GAE/g%. Hasil analisis menunjukkan bahwa kadar fenolik total beras hitam mengalami perubahan signifikan setelah proses hidrolisis. Uji analisa menghasilkan nilai signifikansi sebesar 0,006 (p < 0,05), Peningkatan ini menunjukkan bahwa proses hidrolisis HCl mampu membebaskan senyawa fenolik terikat sehingga kandungan fenolik total meningkat. temuan penelitian ini tidak hanya menambah bukti ilmiah mengenai potensi beras hitam sebagai sumber bioaktif alami, tetapi juga memberikan dasar kuat bagi pengembangannya lebih lanjut ke arah produk fitofarmaka yang terstandarisasi dan teruji secara klinis. Abstract (EN): Black rice (Oryza sativa L. indica) is a pigmented rice rich in phenolic compounds and holds potential as a natural antioxidant. This study aims to determine the total phenolic content in the HCl hydrolyzed water fraction of black rice ethanol extract. This research was conducted from March to July 2025 in the Pharmaceutical Chemistry laboratory. The samples were sourced from Slamet Regency, Central Java. This study used experimental methods with quantitative and qualitative analysis. The extraction results showed a yield of 10.15%. Phytochemical screening identified alkaloids, flavonoids, tannins, phenolics, saponins, steroids, and terpenoids. Fractionation produced an aqueous fraction, which was then hydrolyzed with HCl to release glycones. The total phenolic content was determined using the Folin–Ciocalteu method with UV-Vis spectrophotometry at a wavelength of 730 nm, using gallic acid as a standard. The results showed that the total phenolic content of the water fraction was 73.402 ± 0.173 mg GAE/g %, while that of the ethyl acetate fraction after hydrolysis increased to 91.72 ± 0.420 mg GAE/g%. The analysis results showed that the total phenolic content of black rice underwent a significant change after the hydrolysis process. The analysis test produced a significance value of 0.006 (p < 0.05). This increase indicates that the HCl hydrolysis process was able to release bound phenolic compounds, thereby increasing the total phenolic content. The findings of this study not only add scientific evidence regarding the potential of black rice as a natural bioactive source, but also provide a strong basis for its further development into standardized and clinically tested phytopharmaceutical products.
