Abstrak
Kanker payudara merupakan salah satu penyebab utama kematian pada wanita di Indonesia. Terbatasnya efektivitas dan efek samping dari terapi konvensional mendorong pengembangan alternatif berbasis bahan alam. Salah satu tanaman yang berpotensi sebagai agen antikanker adalah kayu bajakah (Spatholobus littoralis Hassk). Penelitian ini bertujuan untuk mengetahui aktivitas toksisitas ekstrak etanol dan fraksi-fraksi kayu bajakah terhadap larva Artemia salina serta aktivitas antikanker terhadap sel kanker payudara MCF-7. Uji toksisitas dilakukan dengan metode BSLT dan uji aktivitas antikanker dilakukan secara in vitro menggunakan metode WST-8. Hasil uji skrining fitokimia pada ekstrak etanol dan fraksi etil asetat menunjukkan adanya senyawa aktif seperti alkaloid, flavonoid, tanin, terpenoid, steroid, dan saponin. Hasil uji kadar air dan kadar abu ekstrak etanol berturut- turut didapatkan sebesar 8,9% dan 4,28%. Hasil uji toksisitas menunjukkan nilai LC50 dari ekstrak etanol, fraksi n-heksan, fraksi etil asetat dan fraksi etanol berturut sebesar 884 μg/mL, 412 μg/mL, 377 μg/mL, 393 μg/mL. Berdasarkan analisis nilai LC50 ekstrak dan fraksi bersifat toksik. Fraksi etil asetat memiliki nilai LC50 paling rendah sehingga dilanjutkan pada uji sitotoksik terhadap sel kanker payudara MCF- 7. Pada uji sitotoksik fraksi etil asetat mampu menghambat pertumbuhan sel kanker dengan LC50 377 μg/mL dengan kategori cukup aktif. Abstract (EN): Breast cancer is one of the leading causes of death among women in Indonesia. The limited effectiveness and side effects of conventional therapies have driven the development of natural-based alternatives. One plant with potential as an anticancer agent is bajakah wood (Spatholobus littoralis Hassk). This study aims to determine the toxicity activity of ethanol extract and fractions of bajakah wood against Artemia salina larvae and anticancer activity against MCF-7 breast cancer cells. Toxicity testing was conducted using the Brine Shrimp Lethality Test (BSLT) method, while anticancer activity was tested in vitro using the WST-8 method. Phytochemical screening of the ethanol extract and ethyl acetate fraction showed the presence of active compounds such as alkaloids, flavonoids, tannins, terpenoids, steroids, and saponins. The moisture content and ash content of the ethanol extract were found to be 8.9% and 4.28%, respectively. The toxicity test results showed LC50 values for the ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol fraction of 884 μg/mL, 412 μg/mL, 377 μg/mL, and 393 μg/mL, respectively. Based on LC50 analysis, the extract and fractions are considered toxic. The ethyl acetate fraction had the lowest LC50 value and was further tested for cytotoxic activity against MCF-7 breast cancer cells. The cytotoxic test showed that the ethyl acetate fraction could inhibit cancer cell growth with an LC50 value of 377 μg/mL, which falls into the moderately active category.
