Abstrak
Kanker payudara merupakan jenis kanker dengan prevalensi tertinggi pada wanita di dunia. Salah satu tanaman yang berpotensi sebagai agen antikanker adalah kayu bajakah Spatholobus ferrugineus (Zoll. & Moritzi) Benth yang mengandung berbagai metabolit sekunder. Penelitian ini bertujuan mengetahui aktivitas toksisitas ekstrak etanol dan fraksi-fraksi kayu bajakah terhadap larva Artemia salina serta aktivitas antikanker terhadap sel kanker payudara MCF-7. Uji toksisitas dilakukan dengan metode BSLT dan uji aktivitas antikanker dilakukan secara in vitro menggunakan metode WST-8. Hasil uji fitokimia pada ekstrak etanol dan fraksi etil asetat menunjukkan adanya senyawa aktif seperti alkaloid, tanin, flavonoid, terpenoid, steroid dan saponin. Hasil uji kadar air dan kadar abu ekstrak etanol berturut- turut didapatkan sebesar 10% dan 3,6%. Hasil uji toksisitas menunjukkan nilai LC50 dari ekstrak etanol, fraksi n-heksan, fraksi etil asetat dan fraksil etanol berturut-turut adalah 603 μg/mL, 684 μg/mL, 536 μg/mL dan 606 μg/mL. Berdasarkan analisis nilai LC50 semua ekstrak dan fraksi bersifat toksik. Fraksi etil asetat memiliki nilai LC50 yang paling rendah sehingga dilanjutkan pada uji sitotoksik terhadap sel kanker payudara MCF-7. Pada uji sitotoksik fraksi etil asetat mampu menghambat pertumbuhan sel kanker dengan IC50 132 μg/mL dengan kategori sitotoksik moderat atau cukup aktif. Abstract (EN): Breast cancer is the most prevalent type of cancer among women worldwide. One plant with potential as an anticancer agent is Spatholobus ferrugineus (Zoll. & Moritzi) Benth, commonly known as bajakah wood, which contains various secondary metabolites. This study aims to evaluate the toxicity activity of ethanol extract and fractions of bajakah wood against Artemia salina larvae as well as their anticancer activity against MCF-7 breast cancer cells. The toxicity test was conducted using the Brine Shrimp Lethality Test (BSLT) method, while the anticancer activity was tested in vitro using the WST-8 method. Phytochemical screening of the ethanol extract and ethyl acetate fraction revealed the presence of active compounds such as alkaloids, tannins, flavonoids, terpenoids, and saponins. The moisture content and ash content of the ethanol extract were found to be 10% and 3.6%, respectively. Toxicity test results showed LC50 values for the ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol fraction of 603 μg/mL, 684 μg/mL, 536 μg/mL, and 606 μg/mL, respectively. Based on the LC50 values, all extracts and fractions are considered toxic. The ethyl acetate fraction had the lowest LC50 value and was therefore selected for cytotoxic testing against MCF-7 breast cancer cells. In the cytotoxicity test, the ethyl acetate fraction was able to inhibit cancer cell growth with an IC50 value of 132 μg/mL, categorized as moderately cytotoxic or moderately active.
