Abstrak
Kayu kuning sudah lama dikenal dan digunakan secara tradisional untuk pengobatan herbal. Kayu kuning memiliki berbagai khasiat dalam pengobatan seperti dapat menghambat pertumbuhan dan mematikan sel kanker. Penelitian bertujuan menentukan toksisitas dan aktivitas antikanker fraksi kayu kuning. Uji toksisitas dilakukan dengan metode Brine Shrimp Lethality Test (BSLT) dan uji aktivitas antikanker dilakukan secara in vitro menggunakan metode 2-(2-metoksi- 4-nitrofenil)-3-(4-nitrofenil)-5-(2,4-disulfofenil)-2H-tetrazolium (WST-8). Hasil uji fitokimia pada ekstrak etanol dan fraksi etanol menunjukkan adanya senyawa aktif seperti alkaloid, flavonoid, tanin dan steroid. Hasil uji toksisitas menggunakan metode BSLT menunjukkan nilai LC50 masing-masing ekstrak etanol 624,43 ppm, fraksi etanol 552,96 ppm, fraksi etil asetat 950,86 ppm, dan fraksi n-heksan 1095,62 ppm. Berdasarkan analisis nilai LC50 yang kurang dari Abstract (EN): Kayu kuning (Arcangelisia flava (L.) Merr) has long been known and traditionally used as an herbal remedy. It is believed to offer various therapeutic benefits, including the ability to inhibit and kill cancer cells. This study aimed to determine the toxicity and anticancer activity of kayu kuning fractions. Toxicity testing was conducted using the Brine Shrimp Lethality Test (BSLT), while anticancer activity was evaluated in vitro using the WST-8 assay (2-(2-methoxy-4-nitrophenyl)-3-(4- nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium. Phytochemical screening of the ethanol extract and its fractions revealed the presence of active compounds such as alkaloids, flavonoids, tannins, and steroids. BSLT results showed LC5 0 values of 624.43 ppm for the ethanol extract, 552.96 ppm for the ethanol fraction, 950.86 ppm for the ethyl acetate fraction, and 1095.62 ppm for the n-hexane fraction. Based on LC5 0 values, samples with LC5 0 < 1000 ppm are considered toxic. The ethanol fraction exhibited the lowest LC5 0 value and was therefore selected for further anticancer testing. The anticancer activity assay demonstrated that the ethanol fraction inhibited the growth of MCF-7 breast cancer cells with an IC5 0 value of 189.86 ppm, categorizing it as having moderate cytotoxic activity.
