Abstrak
Tanaman daun kelor seperti akar, batang, bunga, buah, biji, dan daunnya dapat dimanfaatkan untuk pengobatan penyakit tertentu. Uji keamanan terhadap obat herbal dilakukan untuk mengetahui dosis aman untuk dikonsumsi Penelitian ini bertujuan untuk mengevaluasi toksisitas akut fraksi etil asetat dari daun kelor (Moringa oleifera Lam.) dan dampaknya terhadap histopatologi hepar pada mencit putih jantan (Mus Musculus). Metode yang digunakan adalah eksperimental dengan desain in vivo. Sebanyak 25 ekor mencit dibagi menjadi lima kelompok, perlakuan dan diberikan fraksi etil asetat daun kelor secara oral dengan dosis bertingkat sesuai metode Thomson dan Weil, yaitu 0 (control), 5 , 50, 500, 5000 mg/kgbb. Pengamatan dilakukan terhadap gejala toksisitas, perubahan berat badan, dan analisis histopatologi hepar setelah 14 hari perlakuan. Hasil menunjukkan bahwa fraksi etil asetat daun kelor tidak menyebabkan kematian pada semua dosis, sehingga nilai LD50 fraksi etil asetat daun kelor diperkirakan lebih dari 5000 mg/kgbb dan kategori tidak toksik. Pemberian fraksi ini berpengaruh signifikan terhadap perubahan berat badan mencit , Hasil analisis histopatologi hepar menunjukkan kerusakan minimal pada dosis tertinggi. Penelitian ini menyimpulkan bahwa fraksi etil asetat daun kelor tidak menimbulkan kematian akut , konsumsi dalam dosis tinggi dapat menyebabkan kerusakan hati signifikan. Abstract (EN): Various parts of the moringa plant—such as the roots, stems, flowers, fruits, seeds, and leaves—can be used to treat certain diseases. Safety testing of herbal medicines is essential to determine safe dosage levels for consumption. This study aims to evaluate the acute toxicity of the ethyl acetate fraction of moringa leaves (Moringa oleifera Lam.) and its effects on the liver histopathology of male white mice (Mus musculus). This research used an experimental in vivo design. A total of 25 mice were divided into five groups and given oral doses of the ethyl acetate fraction in a graded manner according to the Thomson and Weil method: 0 (control), 5, 50, 500, and 5000 mg/kg body weight. Observations were made on toxicity symptoms, body weight changes, and liver histopathology after 14 days of treatment. The results showed that the ethyl acetate fraction of moringa leaves did not cause mortality at any dose, indicating an LD50 value greater than 5000 mg/kg body weight, thus classified as non-toxic. However, administration of the fraction had a significant effect on the body weight of the mice. Histopathological analysis of the liver showed minimal damage at the highest dose. This study concludes that while the ethyl acetate fraction of moringa leaves does not cause acute mortality, high-dose consumption may lead to significant liver damage.
