Abstrak
Radikal bebas adalah molekul reaktif yang dapat merusak lipid, protein, dan DNA sehingga menimbulkan stres oksidatif serta berkontribusi pada penyakit degeneratif, termasuk kanker, diabetes, dan gangguan kardiovaskular. Beras hitam (Oryza sativa L. indica) merupakan sumber alami senyawa bioaktif, seperti fenolik, flavonoid, dan antosianin, yang berpotensi sebagai antioksidan. Penelitian eksperimen dengan analisis kuantitatif ini bertujuan mengetahui kapasitas antioksidan fraksi air hasil hidrolisis HCl dari ekstrak etanol 70% beras hitam menggunakan metode FRAP (Ferric Reducing Antioxidant Power) dan membandingkannya dengan vitamin C sebagai kontrol positif. Penelitian ini dilakukan di Laboratorium Kimia Farmasi Universitas Fort De Kock Bukittinggi. Sampel beras hitam diperoleh dari Kabupaten Sleman, Jawa Tengah. Ekstraksi dilakukan dengan metode maserasi menggunakan etanol 70%, dilanjutkan fraksinasi cair- cair, kemudian hidrolisis HCl untuk memutus ikatan glikosidik sehingga menghasilkan aglikon yang lebih aktif. Hasil penelitian menunjukkan fraksi air terhidrolisis HCl memiliki kapasitas reduksi rata-rata 3,43 ± 2,24 g ekuivalen FeSO4/100 g ekstrak, meningkat menjadi 6,39 mg ekuivalen FeSO4/100 g pada konsentrasi 100 ppm. Nilai tersebut jauh lebih rendah dibandingkan vitamin C (806,22 ± 320,6 g ekuivalen FeSO4/100 g). Meskipun demikian, hidrolisis terbukti meningkatkan kapasitas antioksidan fraksi air beras hitam. Dengan demikian, beras hitam berpotensi dikembangkan sebagai sumber antioksidan alami untuk pencegahan penyakit degeneratif. Abstract (EN): Free radicals are reactive molecules that can damage lipids, proteins, and DNA, leading to oxidative stress and contributing to degenerative diseases such as cancer, diabetes, and cardiovascular disorders. Black rice (Oryza sativa L. indica) is a natural source of bioactive compounds, including phenolics, flavonoids, and anthocyanins, which have potential antioxidant properties. This experimental research with quantitative analysis aimed to determine the antioxidant capacity of the HCl-hydrolyzed water fraction from 70% ethanol extract of black rice using the FRAP (Ferric Reducing Antioxidant Power) method, and to compare it with vitamin C as a positive control. The study was conducted at the Pharmaceutical Chemistry Laboratory, Fort De Kock University, Bukittinggi. Black rice samples were obtained from Sleman Regency, Central Java. Extraction was carried out using maceration with 70% ethanol, followed by liquid-liquid fractionation, and then HCl hydrolysis to break glycosidic bonds, producing more active aglycones. The results showed that the HCl-hydrolyzed water fraction had an average reducing capacity of 3.43 ± 2.24 g FeSO4 equivalent/100 g extract, which increased to 6.39 mg FeSO4 equivalent/100 g at a concentration of 100 ppm. However, this value was significantly lower than that of vitamin C (806.22 ± 320.6 g FeSO4 equivalent/100 g). Nevertheless, hydrolysis was proven to enhance the antioxidant capacity of the water fraction of black rice. Therefore, black rice has potential to be developed as a natural antioxidant source for the prevention of degenerative diseases.
