Abstrak
Kanker payudara merupakan salah satu kanker dengan prevalensi tertinggi. Terapi konvensional seperti kemoterapi sering menimbulkan efek samping pada sel sehat. Penelitian ini bertujuan mengevaluasi aktivitas antikanker ekstrak non polar daun piladang hijau (Coleus monostachyus) terhadap sel kanker MCF-7 secara in vitro serta menentukan pelarut non polar paling efektif dalam mengekstraksi senyawa sitotoksik. Ekstraksi dengan metode maserasi menggunakan pelarut kloroform, diklorometana, dan n-heksan. Kloroform menghasilkan rendemen tertinggi (18,5%), diikuti diklorometana (17,5%) dan n-heksan (15,5%). Skrining fitokimia menunjukkan seluruh ekstrak mengandung alkaloid, flavonoid, tanin, dan steroid. Uji toksisitas metode Brine Shrimp Lethality Test (BSLT) menunjukkan nilai LC50 terendah pada ekstrak kloroform (100,50 μg/mL), diikuti n-heksan (143,05 μg/mL) dan diklorometana (240,71 μg/mL), semuanya tergolong toksik sedang. Uji sitotoksik metode MTT Assay juga menunjukkan IC50 terendah pada ekstrak kloroform (128,82 μg/mL), diikuti n-heksan (150,02 μg/mL) dan diklorometana (278,84 μg/mL). Seluruh ekstrak menunjukkan efek sitotoksik secara konsentrasi- dependen. Hasil menunjukkan bahwa ekstrak daun Coleus monostachyus dengan pelarut kloroform memiliki potensi tertinggi sebagai agen antikanker alami. Penelitian ini mendukung pemanfaatan tanaman obat sebagai alternatif terapi kanker yang lebih aman dan efektif. Abstract (EN): Breast cancer is one of the most prevalent types of cancer globally. Conventional treatments such as chemotherapy often cause adverse side effects on healthy cells. This study aimed to evaluate the anticancer activity of non-polar solvent extracts from green piladang leaves (Coleus monostachyus) against MCF-7 breast cancer cells in vitro, and to determine the most effective non-polar solvent for extracting cytotoxic compounds. Extraction was carried out using the maceration method with three non-polar solvents: chloroform, dichloromethane, and n-hexane. Chloroform yielded the highest extract percentage (18.5%), followed by dichloromethane (17.5%) and n-hexane (15.5%). Phytochemical screening revealed that all extracts contained alkaloids, flavonoids, tannins, and steroids. Toxicity testing using the Brine Shrimp Lethality Test (BSLT) showed the lowest LC50 value in the chloroform extract (100.50 μg/mL), followed by n-hexane (143.05 μg/mL), and dichloromethane (240.71 μg/mL)—all classified as moderately toxic. Cytotoxicity testing via the MTT Assay also showed the lowest IC50 in the chloroform extract (128.82 μg/mL), followed by n-hexane (150.02 μg/mL) and dichloromethane (278.84 μg/mL). These results indicate that the chloroform extract of Coleus monostachyus leaves possesses the strongest potential as a natural anticancer agent. This research supports the potential use of medicinal plants as safer and more effective alternative therapies for cancer treatment.
