Abstrak

Tanaman gambir (Uncaria gambir (W.Hunter). Roxb) memiliki potensi besar dalam pengobatan berbagai infeksi bakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri terhadap Escherichia coli. Metode penelitian meliputi pembuatan ekstrak dan fraksi daun gambir dengan beberapa pelarut, diikuti dengan uji skrining fitokimia dan analisis LC-HRMS. Uji antibakteri dilakukan menggunakan metode difusi cakram pada media Mueller Hinton Agar (MHA) dengan parameter berupa pengukuran diameter zona hambat. Konsentrasi ekstrak yang digunakan adalah 25%, 50%, dan 75%, dengan kloramfenikol sebagai kontrol positif dan DMSO sebagai kontrol negatif. Hasil penelitian menunjukkan ekstrak etanol menghasilkan zona hambat 16,28–22,26 mm (kuat–sangat kuat), sedangkan ekstrak air tidak menunjukkan aktivitas. Fraksi air memberikan daya hambat tertinggi 22,60–26,13 mm (sangat kuat), diikuti fraksi etil asetat 14,52–21,31 mm (kuat–sangat kuat), dan fraksi n-heksan 12,10–17,83 mm (sedang–kuat). Data ini menunjukkan bahwa ekstrak etanol, fraksi etil asetat, fraksi air, dan fraksi n-heksan daun gambir memiliki potensi signifikan dalam menghambat pertumbuhan Escherichia coli. Abstract (EN): The gambir plant (Uncaria gambir (W. Hunter) Roxb.) has significant potential in the treatment of various bacterial infections. This study aimed to evaluate its antibacterial activity against Escherichia coli. The research involved the preparation of gambir leaf extracts and their fractions using various solvents, followed by phytochemical screening and LC-HRMS analysis. Antibacterial activity was assessed using the disk diffusion method on Mueller Hinton Agar (MHA), with the inhibition zone diameter as the main parameter. Extract concentrations tested were 25%, 50%, and 75%, with chloramphenicol as a positive control and DMSO as a negative control. The results showed that the ethanol extract produced inhibition zones ranging from 16.28 to 22.26 mm (indicating strong to very strong activity), whereas the water extract showed no antibacterial activity. Among the fractions tested, the water fraction exhibited the highest inhibitory activity, with inhibition zones of 22.60–26.13 mm (very strong), followed by the ethyl acetate fraction with zones of 14.52–21.31 mm (strong to very strong), and the n-hexane fraction with 12.10–17.83 mm (moderate to strong). These findings demonstrate that the ethanol extract, as well as the water, ethyl acetate, and n-hexane fractions of gambir leaves, possess significant antibacterial potential against Escherichia coli.
