Abstrak

Nanoteknologi berperan penting dalam bidang farmasi, terutama dalam pengembangan nanopartikel logam seperti nanopartikel perak (AgNPs) yang memiliki aktivitas antibakteri, antioksidan, dan antikanker. Penelitian ini bertujuan untuk mensintesis dan mengkarakterisasi AgNPs menggunakan ekstrak etanol dan air daun Gambir (Uncaria gambir (W. Hunter) Roxb). sebagai agen bioreduktor alami serta mengevaluasi aktivitas toksisitasnya menggunakan metode Brine Shrimp Lethality Test (BSLT). Ekstraksi dilakukan dengan metode maserasi (etanol) dan infusa (air), kemudian karakterisasi dilakukan menggunakan LC-HRMS dan Particle Size Analyzer (PSA). Hasil menunjukkan bahwa AgNPs dari ekstrak etanol memiliki ukuran partikel 177 nm, zeta potensial +22,08 mV, dan PDI 0,227, menandakan kestabilan yang baik. Sedangkan ekstrak air menghasilkan partikel 1.333 nm dengan kestabilan lebih rendah. Nilai LC5 0 masing-masing adalah 4,964 μg/mL (etanol) dan 8,44 μg/mL (air), menunjukkan aktivitas toksik kuat terutama pada ekstrak etanol. Abstract (EN): Nanotechnology plays an important role in the pharmaceutical field, particularly in the development of metallic nanoparticles such as silver nanoparticles (AgNPs), which exhibit antibacterial, antioxidant, and anticancer activities. This study aimed to synthesize and characterize AgNPs using ethanolic and aqueous extracts of Uncaria gambir (W. Hunter) Roxb. leaves as natural bioreducing agents and to evaluate their toxicity using the Brine Shrimp Lethality Test (BSLT) method. The extracts were obtained through maceration (ethanol) and infusion (water) methods, then characterized using LC-HRMS and Particle Size Analyzer (PSA). The results showed that AgNPs synthesized from the ethanolic extract had a particle size of 177 nm, a zeta potential of +22.08 mV, and a PDI of 0.227, indicating good stability. Meanwhile, AgNPs from the aqueous extract had a particle size of 1,333 nm, a zeta potential of +7.92 mV, and a PDI of 0.57, indicating lower stability. The LC50 values were 4.964 μg/mL for the ethanolic extract and 8.44 μg/mL for the aqueous extract, demonstrating strong cytotoxic activity, especially in the ethanolic extract.
