Abstrak
Cluster of differentiation-8 (CD8) merupakan marker dari sel T sitotoksik. Sel CD8 sangat penting terhadap penanganan penyakit infeksi virus, kanker/ tumor dan bakteri intraseluler, dan upaya untuk meningkatkan sistem pertahanan tubuh. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian fraksi etil asetat daun alpukat (Persea americana Mill.) terhadap aktivitas sel CD8 sebagai imunomodulator pada tikus putih jantan. Latar belakang penelitian ini didasari oleh pentingnya peningkatan sistem imun. Daun alpukat diketahui mengandung senyawa bioaktif seperti flavonoid, tannin, saponin dan triterpenoid yang berpotensi meningkatkan sistem imun. Penelitian ini menggunakan desain true experimental dengan rancangan pre-post. Tikus dibagi menjadi 3 kelompok, yaitu kelompok kontrol negatif, kelompok pembanding (Stimuno Forte), dan kelompok perlakuan (fraksi etil asetat daun alpukat) dosis 2 ml/200 gr BB yang diberikan secara oral selama 14 hari. Pengukuran kadar CD8 dilakukan dengan metode ELISA sebelum dan sesudah perlakuan. Analisis data menggunakan One-Way ANOVA menunjukkan adanya perbedaan signifikan kadar CD8 antar kelompok (p Abstract (EN): Cluster of Differentiation 8 (CD8) is a marker of cytotoxic T cells. CD8 cells play a crucial role in combating viral infections, cancer/tumors, and intracellular bacterial infections, as well as in efforts to strengthen the body’s immune system. This study aims to determine the effect of the ethyl acetate fraction of avocado leaves (Persea americana Mill.) on CD8 cell activity as an immunomodulator in male white rats. The background of this research is based on the importance of enhancing the immune system. Avocado leaves are known to contain bioactive compounds such as flavonoids, tannins, saponins, and triterpenoids, which have the potential to boost immunity. This study used a true experimental design with a pre-post design. The rats were divided into three groups: a negative control group, a comparison group (Stimuno Forte), and a treatment group (ethyl acetate fraction of avocado leaves) with a dose of 2 ml/200 g body weight, administered orally for 14 days. CD8 levels were measured using the ELISA method before and after treatment. Data analysis using One-Way ANOVA showed a significant difference in CD8 levels among the groups (p < 0.05), where the treatment group showed an increase of 20 cells/μl, comparable to the comparison group with an increase of 22.02 cells/μl. It can be concluded that the ethyl acetate fraction of avocado leaves has potential as a natural immunomodulator through the enhancement of CD8 cell activity.
